The University of Pennsylvania induced Pluripotent Stem Cell Core provides resources for disseminating human pluripotent stem cell technology within UPenn and surrounding research communities. Our services include derivation of patient-specific iPSCs, genome engineering of stem cell lines using CRISPR/Cas9 technology, and lineage specific differentiation of iPSCs/hESCs. The Core has a large collection of patient-derived iPSC lines and cell lines of the gastrointestinal tract (esophageal, pancreatic and intestinal) available to users. The core is also committed to training investigators in stem cell culture techniques and providing quality tested stem cell reagents to users. To initiate a service, please first contact the Core director Wenli Yang to set up a consultation.

iPSC-Derived Cardiomyocytes (each) |
This service provides iPSC-derived cardiomyocytes in monolayer cultures. iPSCs are differentiated using cytokines and small molecules in defined media. Cultures are then metabolically selected to enrich for cardiomyocytes and re-plated. These cells resemble immature cardiomyocytes and but so display a number of properties of adult cardiomyocytes.

iPSC-Derived Hepatocyte Like Cells (HLCs) (each) |
This service provides HLCs in monolayer cultures. iPSCs undergo stage-specific directed differentiation to establish definitive endoderm followed by, hepatic progenitors, immature hepatocytes, and hepatocyte-like cells (HLCs). These cells resemble fetal hepatocytes but do display a number of properties of mature hepatocytes.

iPSC-Derived Endothelial Cells (each) |
This service provides iPSC-derived endothelial cell in monolayer cultures. iPSCs undergo directed differentiation using cytokines and small molecules to endothelial progenitors. Cultures are then purified with endothelial specific cell surface markers to enrich for purity and further passaged. These cells express CD31(PECAM) & CD144 (VE-cadherin) surface markers, uptake Ac-LDL, and form vessels when cultured with stromal cells in 3D cultures.

We provide limited tissue culture space to UPenn investigators who elect to perform their experiments in the iPSC core with guidance from the Core staff. Investigators have full access to our hESC/iPSC dedicated mycoplasma-free cell culture rooms, consumables and some commonly used stem cell culture reagents. All other reagents are charged separately or provided by the user.
